During World War II, obtaining intelligence on other countries' social and economic situations became highly valuable. In response, Japan established a new spy organization called "D Agency" under Lieutenant Colonel Yuuki's command. The agency assigned eight agents to infiltrate and observe powerful countries, reporting on any developments related to the war. To carry out these dangerous missions, the men trained their bodies to withstand extreme conditions and studied various fields such as communications and languages. Their greatest strength was their ability to manipulate people to obtain crucial information that would give Japan an advantage in the conflict.

Joker Game: The Animation is an adaptation of Production I.Gs anime adaptation of Kouji Yanagis novel Joker Game.
